Certainly stance is perhaps the most elementary. Others could be line splits, how far guards or tackles line up off the ball (to facilitate pulling), formations, personnel packages, depth of running backs, sequence of play calling, field position etc.

Generally, we were prohibited from filming as a scouting method period. Often all we had were a pile of papers with plays diagrammed and defensive alignment.
